If the villain's monologue is interrupted do they get surprised?

No.

If neither side tries to be stealthy, they automatically notice each other. Otherwise, the GM compares the Dexterity (Stealth) checks of anyone hiding with the passive Wisdom (Perception) score of each creature on the opposing side.

Interrupting the villain's monologue doesn't give you a surprise round, it results in initiative being rolled. In order to interrupt the villain's speech, you'll need to beat them in the initiative roll. Also, surprise rounds don't exist anymore:

If you're surprised, you can't move or take an action on your first turn of the combat, and you can't take a reaction until that turn ends. A member of a group can be surprised even if the other members aren't.

You only get the Surprised condition if you're snuck up on with a Stealth check, which is made against every enemy in the initiative. The villain's monologue is only interrupted if they're beaten in the initiative roll. However, talking is a free action, so the monologue can continue on their turn. That said, don't give your villains long monologues, most players find them boring (which is why they tried to interrupt it). Instead, they can monologue on their turn in the initiative order, but keep in mind that a turn in combat is 6 seconds, so you should break up the villain's monologue into 6 second chunks on each of their turns.
